Abstract: O feijão (Phaseolus vulgaris) é uma leguminosa que tem um lugar importante na dieta alimentar portuguesa, no consumo fresco e em seco. Do ponto de vista agronómico, é uma espécie importante para incluir nas rotações de culturas, visto possuir vantagens ao nível da fertilidade e da estrutura do solo. No entanto, nos dias de hoje, assiste-se a uma diminuição da produção da cultura de feijão-verde, tanto a nível nacional como a nível mundial. Esta diminuição deve-se às doenças e pragas que afetam a cultura e que levam a uma redução na sua produção. Os nematodes-das-galhas-radiculares (Meloidogyne spp.) e os nemátodes do género Pratylenchus constituem um grave problema na produção da cultura de feijão-verde podendo afetar a qualidade e a produtividade da produção. Uma vez que esta cultura é consumida praticamente em todo o mundo, e em algumas dietas é a fonte principal de proteína, é importante que se adoptem medidas de modo a reverter esse processo. Tendo em conta que o uso de produtos químicos utilizados como meios de proteção contra doenças é bastante dispendioso, nocivo para a saúde e para o ambiente e que nem sempre resolvem por completo os problemas, é importante estudar outros métodos e técnicas eficazes de modo a garantir uma produção de qualidade. A enxertia é uma técnica que tem vindo a ser utilizada em muitos países produtores de culturas hortícolas, incluindo Portugal. Na enxertia recorre-se à utilização de porta-enxertos resistentes ou tolerantes a patogéneos do solo, e nele são enxertadas cultivares comerciais susceptíveis aos patogéneos, mas que possuem as caraterísticas produtivas pretendidas. Consegue-se com esta técnica reduzir os prejuízos causados pelas doenças do solo, e evitar o uso indiscriminado de pesticidas. Neste trabalho pretendemos estudar os efeitos da enxertia das cultivares de feijão-verde Fasili e Oriente em quarto porta-enxertos, nas populações de nemátodes fitoparasitas (Meloidogyne spp. e Pratylenchus spp.), na formação de nódulos de bactérias fixadoras de azoto e na incidência da doença da suberose radicular causada pelo fungo Pyrenochaeta lycopersici. Pretendemos ainda avaliar o efeito da enxertia na produção de biomassa de raízes de feijão-verde produzido em estufa. O ensaio experimental decorreu no solo da estufa da Escola Superior Agrária de Ponte de Lima/IPVC, na Primavera/Verão de 2014. Realizaram-se duas recolhas de raízes para análise, uma a meio do ciclo da cultura a 1 de Julho e uma segunda no final da cultura em Agosto de 2014. Quantificou-se o número de galhas de nemátodes Meloidogyne spp. (nemátode das galhas radiculares - NGR) e o número de massas de ovos, o número de nemátodes Pratylenchus spp. extraídos das raízes mais finas, o número de nódulos de rizóbios, e a severidade da doença da suberose radicular. Registou-se ainda o peso fresco e seco das raízes de feijoeiro das diferentes combinações porta-enxerto/cultivar enxertada. A partir dos nódulos de rizóbio, isolaram-se as bactérias em laboratório e procedeu-se à caraterização fenotípica e molecular. O estudo dos nemátodes fitoparasitas presentes nas suspensões do solo, permitiram identificar um total de sete géneros de nemátodes nos vários tratamentos e datas de amostragem do ensaio: os ectoparasitas Helicotylenchus, Paratylenchus, Rotylenchus, Tylenchorhynchus e Xiphinema; e os endoparasitas Meloidogyne e Pratylenchus. Os resultados mostraram que as populações naturais de nemátodes, rizóbio e P. lycopersici presentes no solo infetaram as raízes dos porta-enxertos em estudo. Nenhum dos tratamentos realizados (porta-enxerto/cultivar enxertada) mostrou ser simultaneamente resistente aos nemátodes dos géneros Meloidogyne e Pratylenchus. O tratamento que conduziu a melhores resultados contra Meloidogyne spp. e P. lycopersici, foi o tratamento OO (Oriente enxertado em Oriente), enquanto que para o género Pratylenchus o tratamento mais aconselhável foi o FF (Fasili enxertado em Fasili). No que diz respeito à infeção por rizóbio não pareceu haver uma separação entre os tratamentos de plantas enxertadas em si próprias ou as plantas não enxertadas. A análise da diversidade genética dos isolados de Rizobium obtidos das raízes de plantas da cv. Feijão de 7 anos (P. coccineus), uma espécie de feijoeiro diferente das cvs. Fasili e Oriente (P. vulgaris), mostrou que as bactérias que infetaram estas duas espécies de feijão são diferentes, e não ficaram agrupadas no mesmo grupo.
The bean (Phaseolus vulgaris) is a legume that has an important place in the Portuguese diet, whether eaten dry or fresh. From an agronomic point of view, it is an important species to include in crop rotations, as it possesses advantages in terms of fertility and soil structure. However, today, we are witnessing a decrease in the production of green beans culture, both nationally and globally. This decrease is due to diseases and pests affecting the crop, leading to lower production. Root-knot nematodes (Meloidogyne spp.) and gender Pratylenchus nematodes are a serious problem in the production of green beans culture and can affect the quality and productivity of production. Since this culture is consumed almost worldwide, and in some diets is the main source of protein, it is important to take measures to reverse the process. Given that the use of chemical products used as protection against disease means is quite expensive, harmful to health and the environment and that does not always solve completely the problems, it is important to study other effective methods and techniques to ensure the quality of production. Grafting is a technique that has been used in many countries producing horticultural crops, including Portugal. Grafting refers to the use rootstock resistant or tolerant to soil pathogens and there are grafted cultivars susceptible to pathogens, but which have the desired characteristics production. With this technique one is able reduce the damage caused by soil-borne diseases, and avoid the indiscriminate use of pesticides. In this work we intend to study the effects of grafting of green beans cultivars Fasili and East fourth rootstocks, in populations of plant parasites nematodes (Meloidogyne spp. and Pratylenchus spp.), in the formation of nitrogen fixing bacteria nodules and incidence corky root of disease caused by the fungus Pyrenochaeta lycopersici. We also intend to evaluate the effect of grafting on the biomass of roots green beans produced in greenhouse. The experimental test took place on the soil of greenhouse Agricultural School of Ponte de Lima / IPVC in spring / summer 2014. There were two collections of roots for analysis, one half of the cycle on 1 July and a second at the end of culture in August 2014. Quantified the number of galls of nematodes Meloidogyne spp. (Root-knot nematodes-RKN), and the number of egg masses, the number of nematode Pratylenchus spp. extracted the fine roots, the number of rhizobia nodes, and the severity of corky root disease. There has still fresh and dry weight of the bean roots of different combinations rootstock / cultivar grafted. From the rhizobia nodules were isolated bacteria in laboratory and proceeded to phenotypic and molecular characterization. The present study of phytoparasitic nematodes in soil suspensions, identified a total of seven genera of nematodes in the various treatments and test sampling dates: ectoparasites Helicotylenchus, Paratylenchus, Rotylenchus, Xiphinema and Tylenchorhynchus; and endoparasites Meloidogyne and Pratylenchus. The results showed that the natural populations of nematodes, rhizobia and P. lycopersici in the soil infect only the roots of the rootstocks under study. None of the treatments (rootstock / cultivar grafted) proved to be simultaneously resistant to nematodes of Meloidogyne and Pratylenchus genera. The treatment led to better results against Meloidogyne spp. and P. lycopersici was OO treatment (Middle East grafted), while for the genus Pratylenchus most advisable treatment was FF (Fasili grafted Fasili). With regard to infection by rhizobia, does not appear to be a separation between treatments grafted plants or the plants themselves ungrafted. Analysis of genetic diversity of Rhizobium isolates obtained from cv. plant roots. Bean 7 years (P. coccineus), a different kind of bean from cvs. Fasili and East (P. vulgaris), showed that the bacteria that infect only these two bean species are different, and were not grouped in the same group.
