Title: Prevalência e caracterização de Escherichia Coli Stec em fezes de vaca de produção de leite

Abstract: A realização deste trabalho teve como principal objetivo estudar a presença da bactéria patogénica Escherichia coli produtoras de toxinas shiga (stx) em explorações de vacas leiteiras e avaliar sua resistência a antibióticos. Foram acompanhados um total de 80 animais distribuídos por 4 explorações. Em três das explorações (A, C e D) foram recolhidas amostras de fezes de vacas lactantes e em uma (B) foram recolhidas amostras de fezes de novilhas. A metodologia utilizada para a deteção de STEC foi a constante na ISO/TS 13136:2012, que estabelece a pesquisa dos genes stx1, stx2 e eae no meio de enriquecimento da amostra. No caso de resultados positivos procede-se ao isolamento do microrganismo em meios seletivos. O documento estabelece, igualmente, a determinação dos principais serotipos responsáveis por infeção no Homem. Os isolados encontrados foram conservados a -80 ºC. Posteriormente, foi determinada a concentração mínima inibitória (MIC) pelo método da microdiluição. Os dados foram analisados em Microsoft® Office Excel versão 2010. Em 80 amostras recolhidas e analisadas 54 (67,5%) possuíam o gene stx1/stx2 e 16 (20%) o gene eae. Em 31 amostras foi possível determinar o serotipo. A exploração com maior percentagem de animais colonizados é a B (100%) e a A (40%) é a que possuiu menos. O gene de virulência mais prevalente foi o stx2 e o serotipo mais comum foi o O145 não tendo sido encontrado nenhum O111. Nas pools apenas 26,3% mostraram genes de virulência, sendo novamente a exploração B a que apresentou maior prevalência. No fim foram conservadas 23 estirpes STEC e 61 não STEC. Realizando-se de seguida a suas resistências a agentes antimicrobianos foi possível verificar que a ampicilina é o antibiótico com maior número de isolados que apresentaram resistência, seguida da tetraciclina e da amoxicilina. A exploração em que os isolados (STEC e não STEC) apresentaram maior resistência aos antibióticos foi a B.
The main objective of this work was to study the presence of the pathogenic bacteria Shiga Toxins (stx) producing Escherichia coli in dairy farms, and to evaluate their resistance to antibiotics. A total of 80 animals were sampled from 4 farms. The faeces samples of 3 farms (A, C e D) were collected from suckling cows and faeces samples from the other farm (B) were collected from heifers. The methodology used for the detection and identification of STEC followed the ISO/TS 13136:2012, which establishes the search of stx1, stx2 and eae genes from samples enrichment growth. In the case of positive results, microorganisms were isolated using a selective media. The document also establishes the search for the main serotypes responsible for human infection in Europe. The isolates detected were stored at -80 °C. Additionally, the minimum inhibitory concentration (MIC) was determined by the microdilution method. The data were analyzed in Microsoft® Office Excel version 2010. In the 80 samples collected and analyzed 54 (67.5%) have the gene stx1/stx2 and 16 (20%) the gene eae. In 31 of the samples was possible to identified one or more serotypes. The holding with the highest percentage of colonize animals is B (100%) and A (40%) have the lower percentage.The virulence gene most prevalent is stx2 and the most common serotype is O145, in contrast there was none O111 found. Only 26.3 % of the pools had virulence genes detected, with the most prevalent being from farm B. In total 23 STEC and 61 non-STEC strains were isolated and conserved. The exploration in which the isolates (STEC and non-STEC) show higher resistance to antibiotics is B Ampicillin is the antibiotic to which the majority of isolates presented resistance, followed by tetracycline and amoxicillin.
